Expression of cellular prion protein ( PrPc ) in the cat central nervous system . Some findings

چکیده
The expression of cellular prion protein in the central nervous system of normal cats and in cases of kainic acid injections into the mediodorsal thalamic nucleus were studied. Cellular prion protein immunodetection varied in a rostrocaudal direction: the protein was less abundant in the brainstem than in the prosencephalon. Especially abundant were the positive cells in the cerebral cortex. The immunodetection was present in areas projecting to the intermediate band of the thalamic mediodorsal nucleus and to the anteroventral nucleus of the thalamus. Some of these cells were situated around vessels in the basal prosencephalon. The immunodetection was less abundant in cases of kainic acid lesions. NAPPH-diaphorase activity was also studied. In control cases, we observed positive NADPHdiaphorase cells around vessels, especially in the basal prosencephalon. The amounts of such cells and the intensity of the reaction was lower after kainic acid lesions. Our findings lead us to propose a retrograde propagation of altered prion proteins in human spongiform encephalopathies such as fatal familial insomnia; a possible alteration of nitrergic systems is possible. Moreover, there is possibly an alteration of the prion and nitrergic physiology simply due to the destruction of the thalamic mediodorsal nucleus in fatal familial insomnia.

Mechanistic prospective for human PrPC conversion to PrPSc: Molecular dynamic insights
PrPC conversion to PrPSc isoform is the main known cause for prion diseases including Crutzfeldt-Jakob, Gerstmann-Sträussler-Sheinker syndrome and fatal familial insomnia in human. The precise mechanism underling this conversion is yet to be well understood. In the present work, using the coordinate file of PrPC (available on the Protein Data Bank) as a starting structure, separate molecular d...
متن کاملA Study on The Effect of Temperature on Human Prion Protein Structure through Molecular Dynamic Simulation
Background & Aims: The normal form of the prion protein is called PrPC and its infectious form is called PrPSc. This protein functions like a crystallized core for the transformation of PrPc into an abnormal PrPSc. The aim of the present study was to investigate the effect of temperature on human prion protein structure through molecular dynamic simulation. Methods: In this research, the GROMAC...
متن کاملDo Bovine Lymphocytes Express a Peculiar Prion Protein?
The cellular prion protein (PrPc) is a glycolipid-anchored cell surface protein that usually exhibits three glycosylation states. Its post-translationally modified isoform, PrPsc, is involved in the pathogenesis of various transmissible spongiform encephalopathies (TSEs). In bovine species, BSE infectivity appears to be restricted to the central nervous system; few or no detectable infectivity ...
متن کاملMyositis facilitates preclinical accumulation of pathological prion protein in muscle
BACKGROUND In human and animal prion diseases, pathological prion protein, PrPSc, as well as prion infectivity is mainly found in the central nervous system, but also in lymphoid organs and muscle. Pathophysiology of prion colonization of lymphoid organs has been studied intensively, yet how myositis influences prion accumulation in muscle is unknown. RESULT We have investigated the influence...
متن کاملMolecular mechanisms of prion pathogenesis.
Prion diseases are infectious neurodegenerative diseases occurring in humans and animals with an invariably lethal outcome. One fundamental mechanistic event in prion diseases is the aggregation of aberrantly folded prion protein into large amyloid plaques and fibrous structures associated with neurodegeneration. The cellular prion protein (PrPC) is absolutely required for disease development, ...
